How do Phosphorylation ELISA Kits work?
This Sandwich enzyme - linked immunosorbent assay (ELISA) kit is an in vitro assay for the measurement of phosphorylated protein. The 96 - well plate is coated with a specific capture antibody (anti-pan-antibody or anti-phosphorylated antibody) in which sample is pipetted into the wells and the immobilized antibody will bound any target protein present in the sample to the sides of the wells. The wells are then washed. An anti - phosphorylated antibody detects the target protein. A secondary antibody is then pipetted into the wells and they are then washed again. A TMB substrate solution is added to the wells and this promotes colour development proportional to the amount of target bound to the wells. A stop solution changes the colour of the target protein from blue to yellow and the colour intensity in measured at 450nm.
