The ACOT1 Antibody (PAC056406) is a polyclonal antibody designed for research involving ACOT1, an enzyme involved in lipid metabolism. This antibody, raised in rabbits, is highly reactive with human samples and is validated for use in various applications, including Western blot and immunofluorescence. ACOT1 plays a critical role in the hydrolysis of acyl-CoA esters, making it essential for fatty acid metabolism. Its dysregulation has been linked to various metabolic disorders, making it a valuable target for research in areas such as obesity, diabetes, and cardiovascular disease.
By using the ACOT1 Antibody, researchers can detect and analyze the expression of ACOT1 in different cell types and tissues, aiding in the understanding of its functions and potential implications in disease. This antibody is a valuable tool for studies in lipid metabolism, metabolic diseases, and drug discovery efforts aimed at targeting ACOT1 for therapeutic purposes.
IHC image of PACO56406 diluted at 1:300 and staining in paraffin-embedded human kidney tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
Immunofluorescence staining of Hela cells with PACO56406 at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
Background:
Acyl-CoA thioesterases are a group of enzymes that catalyze the hydrolysis of acyl-CoAs to the free fatty acid, and coenzyme A (CoASH), providing the potential to regulate intracellular levels of acyl-CoAs, free fatty acid, and CoASH. Active towards fatty acyl-CoA with chain-lengths of C12-C16 (By similarity).
