Description
UBR1 Antibody (PACO56414)
The UBR1 Polyclonal Antibody (PAC056414) is a valuable tool for researchers studying UBR1, a protein involved in ubiquitination and protein degradation processes. This antibody, produced in rabbits, exhibits high reactivity with human samples and is validated for use in Western blot applications. By binding specifically to UBR1, the antibody enables accurate detection and analysis of this protein in a variety of cell types, making it an excellent choice for studies in molecular biology and cellular physiology.UBR1 plays a crucial role in regulating the turnover of key proteins within cells, impacting various cellular processes such as cell cycle progression, DNA repair, and signal transduction.
Dysregulation of UBR1 function has been implicated in diseases like cancer, neurodegenerative disorders, and metabolic conditions, highlighting the importance of understanding its mechanism of action. Research into UBR1 may lead to the development of targeted therapies for these diseases by manipulating protein degradation pathways.

IHC image of PACO56414 diluted at 1:500 and staining in paraffin-embedded human pancreatic tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. | |
Immunofluorescence staining of U251 cells with PACO56414 at 1:166, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
Background: | E3 ubiquitin-protein ligase which is a component of the N-end rule pathway. Recognizes and binds to proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their ubiquitination and subsequent degradation. May be involved in pancreatic homeostasis. Binds leucine and is a negative regulator of the leucine-mTOR signaling pathway, thereby controlling cell growth. |
Synonyms: | E3 ubiquitin-protein ligase UBR1 (EC 2.3.2.27) (N-recognin-1) (RING-type E3 ubiquitin transferase UBR1) (Ubiquitin-protein ligase E3-alpha-1) (Ubiquitin-protein ligase E3-alpha-I), UBR1 |
UniProt Protein Function: | UBR1: E3 ubiquitin-protein ligase which is a component of the N-end rule pathway. Defects in UBR1 are a cause of Johanson-Blizzard syndrome (JBS). This disorder includes congenital exocrine pancreatic insufficiency, multiple malformations such as nasal wing aplasia, and frequent mental retardation. Pancreas of individuals with JBS do not express UBR1 and show intrauterine- onset destructive pancreatitis. Belongs to the UBR1 family. 2 isoforms of the human protein are produced by alternative splicing. |

NCBI Summary: | The N-end rule pathway is one proteolytic pathway of the ubiquitin system. The recognition component of this pathway, encoded by this gene, binds to a destabilizing N-terminal residue of a substrate protein and participates in the formation of a substrate-linked multiubiquitin chain. This leads to the eventual degradation of the substrate protein. The protein described in this record has a RING-type zinc finger and a UBR-type zinc finger. Mutations in this gene have been associated with Johanson-Blizzard syndrome. [provided by RefSeq, Jul 2008] |
