What is Tau?
Tau as a biomarker
There have been many different tau alterations observed in neurodegenerative disorders. At the gene level there are mutations which can lead to rare familial dementia. At the mRNA level there are alterations in alternative splicing which results in abnormal forms of tau isoforms. Lastly, at the protein level there is abnormal hyperphosphorylation which causes aggregation of abnormal tau (p-tau). This altered phosphorylation results in a loss in microtubule stability and alterations in cellular integrity and cytoskeletal maintenance.
Tauopathies are a set of neurodegenerative disorders which are associated with this aggregation of abnormal tau. Some examples of tauopathies are Alzheimer’s disease, corticobasal syndrome and progressive supranuclear palsy. When tau inclusions form in the brain during tauopathies, abnormal tau leaks into the CSF which can be later quantified. The biochemical characterization of tau inclusions has allowed for an improved diagnosis of the aforementioned neurodegenerative diseases and it has shown to have potential as a diagnostic biomarker.
