USP46 belongs to the peptidase C19 family; USP12/USP46 subfamily. Deubiquitinating enzymes (DUBs) are a large group of proteases which are also commonly referred to as deubiquitinating peptidases; deubiquitinating isopeptidases; deubiquitinases; ubiquitin proteases; ubiquitin hydrolyases; ubiquitin isopeptidases; or Dubs. By cleaving ubiquitin-protein bonds, they regulate ubiquitin-dependent metabolic pathways. They may also be used to regulate the ubiquitin system in a negative or positive manner. They are also involved in the processing of ubiquitin precursors, protein ubiquitination, and the dismantling of inhibitory ubiquitin chains. The Usp46 protein is a deubiquitinating enzyme that has an effect on behavior; it's possible that it controls GABA activity. It may help to regulate GAD1/GAD67 deubiquitination by acting as a modulator. The enzyme USP46 has little deubiquitinating activity on its own, and it requires the assistance of WDR48 to be active, which suggests that it is not involved in monoubiquitinated FANCD2 deubiquitination.
