KIR2DL4, also known as CD158d, is a member of the killer cell Ig-like receptor (KIR) family. KIRs are transmembrane glycoproteins expressed by natural killer cells and subsets of T cells. The KIR genes are polymorphic and highly homologous. NK cells and certain T cells contain the gene KIR2DL4. The KIR2DL4 protein activates the phagocytosis of NK cells, even though it contains an immunoreceptor tyrosine-based inhibition motif (ITIM) in its cytosolic tail. The ITIM was not required for KIR2DL4-induced lysis. KIR2DL4's activation signal was diminished by the blockade of another ITIM-containing receptor. KIR2DL4 activation-deficient molecules inhibited the CD16 signal, which was confirmed by blocking the signaling pathway.
