ISG15 is a 15-kDa protein of unique primary amino acid sequence that functions intracellularly as an ubiquitin homologue and a cytokine that stimulates IFN-gamma production and enhances NK / lymphokine-activated killer cell proliferation and function. ISG15 is released by monocytes and lymphocytes. ISG15 is a ubiquitin-like protein that increases as a primary response to various microbial and cellular stressors. The ISG15 signalling pathway has been disrupted in several human tumour types. ISG15 is also induced by viral or bacterial infection via the Janus kinase/signal transducer and activator of transcription (Jak/STAT) signaling pathway, which is known to be activated by type I interferon. ISG15 is released into the circulation by monocytes, B- and T-lymphocytes, and fibroblasts following induction. We show that the function of the ISG15 protein is blocked by influenza B virus, which strongly promotes it. A certain region of the influenza B virus NS1 protein, which includes part of its effector domain, prevents the covalent linkage between ISG15 and its target proteins both in vitro and in infected cells.
