Interleukin-4 (IL-4) is a pleiotropic cytokine that controls a variety of T and B cell activities, including cell growth, survival, and gene expression. T cells, mast cells, and bone marrow stromal cells all make IL-4. IL-4 modulates the differentiation of naive CD4+ T cells into helper Th2 cells; characterized by a cytokine-release profile that includes IL-4, IL-5, IL-6, and IL-13; which promotes a humoral immune response. IL-4 has a major influence on the switching of immunoglobulins to the IgG1 and IgE isotypes. Excessive IL-4 production by Th2 cells is linked to excessive IgE synthesis and an allergic reaction.
