Interleukin 31 (IL-31) is a cytokine with a four-helix bundle structure. It has structural and functional similarities to IL-6, Oncostatin M, LIF, and Cardiotrophin-1. The human IL-31 gene encodes a precursor of 164 amino acids that includes a 23 amino acid signal peptide and a 141 amino acid mature protein. IL-31 has human and murine homologues with a sequence identity of 24 percent in the mature region. IL-31 is mainly associated with activated T cells and is preferentially expressed by type 2 helper T cells (Th2). The IL-31 signaling pathway is activated by the heterodimeric receptor complex consisting of an IL-31RA (also known as GPL and GLMR) protein and an Oncostatin M receptor (OSM R-beta). Keratinocytes express the IL-31 receptor in a permanent manner and can be induced by IFN gamma on monocytes. GPL/OSMR signaling is a strong activator of STAT3 and STAT5, and can also activate STAT1, Jak1, and Jak2 signaling pathways. IL-31 regulates immune responses that are linked with skin physiology and inflammatory skin diseases. Studies have shown that IL31 induces severe pruritis (itching) and dermatitis in transgenic mice.
