Interleukin-22 (IL-22) is a member of the IL-10 family, a group of powerful inflammatory mediators. DC and T cells are responsible for the synthesis of IL-22. The IL-22 and IL-10 receptor chains assist in cellular targeting and signal transduction. It can stimulate and regulate innate immune responses, especially in epithelial cells such as respiratory and intestinal epithelial cells, against bacterial pathogens. The IL-22 cytokine is involved in the coordinated response of both adaptive and innate immune systems. IL-22 has been shown to stimulate hepatocyte survival in the liver and epithelial cells in the lung and gastrointestinal tract, comparable to that of IL-10. IL-22 causes biological activity by interacting with a cell-surface complex that includes IL-22R1 and IL-10R2 receptor chains. IL-22 binds to a soluble binding protein, IL-22BP, before it can interact with other cytokines and activate the cell. IL-22BP and an extracellular region of IL-22R1 share sequence similarity. The pro-inflammatory versus tissue-protective activities of IL-22 are regulated by cytokine IL-17A in some situations.
