Interleukin 8 (IL-8), also known as CXCL8, a chemokine with a distinctive CXC amino acid motif that was originally characterized for its leukocyte chemotactic activity, is now recognized to have tumorigenic and proangiogenic effects. In response to inflammatory signals, this chemokine is produced by various cell types including monocytes/macrophages, T cells, neutrophils, fibroblasts, endothelial cells, and various tumor cell lines. IL-8 is highly expressed and secreted in gliomas in both culture and in animals, and it appears to be important for glial tumor neovascularity as well as tumor growth. Levels of IL-8 indicate glial neoplasms' histological grade, with the most malignant kind, glioblastoma, exhibiting the highest expression in pseudopalisading cells surrounding necrosis, suggesting that hypoxia/anoxia may promote gene transcription. Interleukin (IL) -8/CXCL8 is a strong neutrophil chemotactic factor. Various cell types can release a large amount of IL-8/CXCL8 in response to a wide range of stimuli, including proinflammatory cytokines, microorganisms and their byproducts, as well as environmental changes such as hypoxia, reperfusion, and hyperoxia. IL-8/CXCL8 has been identified as a crucial mediator in neutrophil-mediated acute inflammation, especially owing to its significant influence on neutrophils. However, there is a wealth of evidence that IL-8/CXCL8 has immunomodulatory effects on various cell types, including lymphocytes, monocytes, endothelial cells, and fibroblasts, in addition to neutrophils. These biological activities suggest that IL-8/CXCL8 has important functions in a variety of disease processes, including chronic inflammation and cancer. IL-8 has been linked to tumor blood vessel growth, metastasis, and poor prognosis in breast cancer. IL-8 may present a novel therapeutic target for estrogen driven breast carcinogenesis and tumor progression.
