The histones are a large, complex family of highly conserved basic proteins that bind chromosomal DNA into nucleosomes. There are numerous subtype diversities: H1, H2A, H2B, and H3 or H4. Histone modifications have increasingly been shown to be key players in the control of chromatin states and dynamics as well as gene expression. As a result, histone modifications and enzymatic machineries that establish them are key regulators of cell growth, differentiation, flexibility, and malignancy. However, extracellular histones are a double-edged sword since they may also harm host tissue and cause death. Histones bound to platelets, induced calcium influx, and recruited plasma adhesion proteins such as fibrinogen to induce platelet aggregation. The protein H2a, also known as histone H2A (HIST3H2A), is a component of histones. Covalent proteolysis and modification of histones is essential for chromatin dynamics and transcription. One example of such modification is ubiquitination, which mainly occurs on histones H2A and H2B. E3 ubiquitin ligase complex is specific for histone H2A (HIST3H2A). In HeLa cells, reducing the expression of Ring2 promotes ubiquitination and degradation of H2A. DNA damage induces monoubiquitylation of histone H2A (HIST3H2A) in the vicinity of DNA lesions.
