Endothelial cell-selective adhesion molecule (ESAM) is a JAM family of immunoglobulin superfamily members that includes one V-type and one C2-type immunoglobulin domain, as well as a hydrophobic signal sequence, a single transmembrane region, and a cytoplasmic. It is particularly concentrated in the endothelium and on activated platelets. ESAM at endothelial tight junctions aids neutrophil migration across the vessel wall; it's possible that it does so by influencing endothelial cell contacts. The adaptor protein membrane-associated guanylate kinase MAGI-1 has been shown to bind ESAM. ESAM has previously been shown to influence the development of primary tumour growth and endothelial permeability. ESAM appears to play a functional redundancy in physiological angiogenesis, yet it has a distinct and important function in pathological angiogenic processes such as tumour development.
