Calcium/Calmodulin-Dependent Protein Kinases, or CaM Kinases for short, are a family of serine/threonine-specific protein kinases that are primarily controlled by the Calcium/calmodulin system. CaM kinases exhibit a memory effect. The activity of CaM kinases can outlast the intracellular calcium spike required to activate it. This property is essential for the induction of synaptic plasticity in neurons. The induction of long-term potentiation is blocked by pharmacological inhibition of CaM kinases II. CaM kinases II phosphorylates postsynaptic glutamate receptors following stimulation, altering the synapse's electrical properties. A calcium/calmodulin-dependent protein kinase type 1D, also known as CaM kinase I delta, CaM kinase ID, CaMKI-like protein kinase, CKLiK and CAMK1D, is a member of the protein kinase superfamily and CaMK subfamily. It contains one protein kinase domain. CAMK1D is broadly expressed. The protein is highly and mostly expressed in polymorphonuclear leukocytes (neutrophilic and eosinophilic granulocytes), with minimal or no expression seen in monocytes and lymphocytes. Overexpression of CAMK1D in non-tumorigenic breast epithelial cells resulted in increased cell proliferation, as well as molecular and phenotypic changes indicative of epithelial-mesenchymal transition (EMT), such as loss of cell-cell adhesions and enhanced cell migration and invasion. CAMK1D is a potential therapeutic target with particular relevance to clinically unfavourable basal-like tumors.
