B lymphocyte stimulator (BLyS); also known as TNFSF13B; CD257 and BAFF; is single-pass type II membrane protein; which belongs to the tumour necrosis factor family. BAFF is abundantly expressed in peripheral blood Leukocytes and is especially present in monocytes and macrophages. BAFF is a cytokine that binds to receptors TNFRSF13B (TACI), TNFRSF17 (BCMA), and TNFRSEF13C (BAFFR). B-cell development, homeostasis, and selection are all influenced by these receptors. BLyS levels affect survival signals and selective apoptosis of autoantibody-producing B cells. BAFF is a powerful B-cell activator that has been shown to play an essential part in the multiplication and differentiation of B cells. Overexpression of BLyS in mice induces clinical and serological features of systemic lupus erythematosus (SLE) and Sjogren's syndrome (SS). In human rheumatic diseases, BLyS is a potential drug target. The capacity of BLyS to control both size and repertoire in the peripheral B cell compartment raises the prospect that BLyS and its adversers might serve as a therapeutic trichotomy. As an agonist, BLS protein may improve humoral immunity in individuals with congenital or acquired immunodeficiency as a result of viral infection or cancer therapy.
