Adenylate kinase isoenzyme 4; mitochondrial; also known as ATP-AMP transphosphorylase; Adenylate kinase 3-like; AK4 and AK3L1; is a member the adenylate kinase family. AK4 / AK3L1 is localized to the mitochondrial matrix. Adenylate kinases control the adenine and guanine nucleotide compositions in a cell by allowing for the transfer of phosphate groups among these components. In vertebrates, five isozymes of adenylate kinase have been discovered. Isozymal variants of these enzymes are tissue-specific and developmentally controlled. AK4/AK3L1 is a kinase that participates in the irreversible transfer of the final phosphate group from ATP to AMP. It may also be involved in GTP production. Although adenylate kinase 4 (AK4/AK3L1) is a unique individual that has no enzymatic activity in the adenylate kinase (AK) family, it shows high sequence similarity with other AKs. It's unclear what physiological function AK4 serves and why it is enzymatically inactivated. It is also notable that the AK4 /AK3L1 variant retains the ability to bind nucleotides. It has glutamine instead of an arginine molecule in the active site. The enzymatically inactive AK4 is a stress-responsive protein that is necessary for cell survival and growth. AK4 / AK3L1 is likely to require interaction with the mitochondrial inner membrane protein ANT in order for it to provide protection to cells under stress. AK4 / AK3L1 also acts on the specific mechanism of energy metabolism rather than control of the homeostasis of the ADP pool ubiquitously.
