Description
JAK1 (Phospho-Tyr1022)Colorimetric Cell-Based ELISA Kit
The JAK1 Phospho-Tyr1022 Colorimetric Cell-Based ELISA Kit is designed for the accurate detection of phosphorylated JAK1 (Janus kinase 1) levels in cell lysates. This kit offers high sensitivity and specificity, ensuring precise and reproducible results for research applications involving signaling pathways and cell activation.Phosphorylation of JAK1 at tyrosine 1022 is a key event in the JAK-STAT signaling pathway, which plays a critical role in cell growth, differentiation, and immune response. Dysregulation of this pathway is implicated in various diseases, including cancer, autoimmune disorders, and inflammatory conditions.
The ability to measure phosphorylated JAK1 levels provides valuable insights into these diseases and potential therapeutic targets. The JAK1 Phospho-Tyr1022 Colorimetric Cell-Based ELISA Kit is a valuable tool for researchers studying intracellular signaling pathways and exploring novel therapeutic interventions. The JAK1 (Phospho-Tyr1022) Colorimetric Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can detect JAK1 protein phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated JAK1 in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on JAK1 phosphorylation.
Qualitative determination of JAK1 (Phospho-Tyr1022) concentration is achieved by an indirect ELISA format. In essence, JAK1 (Phospho-Tyr1022) is captured by JAK1 (Phospho-Tyr1022)-specific primary antibodies while the HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Through this binding, the HRP enzyme conjugated to the secondary antibody can catalyze a colorimetric reaction upon substrate addition. Due to the qualitative nature of the Cell-Based ELISA, multiple normalization methods are needed:
1. | A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. |
2. | Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density. After staining, the results can be analysed by normalizing the absorbance values to cell amounts, by which the plating difference can be adjusted. |

UniProt Protein Function: | JAK1: a widely expressed non-receptor tyrosine-kinase involved in the interferon-alpha/beta and -gamma signal transduction pathways. Couples cytokine ligand binding to tyrosine phosphorylation of various known signaling proteins and of a unique family of transcription factors termed the signal transducers and activators of transcription, or STATs. A selective Jak1 inhibitor induces apoptosis in NRP-154 prostate cancer cell line. A single mutation seen in tyrosine kinome screen of colon cancers. Activated in B cell lines from patients with post-transplant lymphoproliferative disorder, and in a mouse Lck-driven T cell lymphoma. Inhibitor: Piceatannol. |

NCBI Summary: | This gene encodes a membrane protein that is a member of a class of protein-tyrosine kinases (PTK) characterized by the presence of a second phosphotransferase-related domain immediately N-terminal to the PTK domain. The encoded kinase phosphorylates STAT proteins (signal transducers and activators of transcription) and plays a key role in interferon-alpha/beta and interferon-gamma signal transduction. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Mar 2016] |

Step | Procedure |
1. | Seed 200 µL of 20,000 adherent cells in culture medium in each well of a 96-well plate. The plates included in the kit are sterile and treated for cell culture. For suspension cells and loosely attached cells, coat the plates with 100 µL of 10 µg/ml Poly-L-Lysine (not included) to each well of a 96-well plate for 30 minutes at 37°C prior to adding cells. |
2. | Incubate the cells for overnight at 37°C, 5% CO2. |
