Glutaminyl-Peptide Cyclotransferase, also known as QPCT is a member ofthe glutaminyl-peptide cyclotransferase family. QPCT is responsible for thebiosynthesis of pyroglutamyl peptides. Furthermore, QPCT is partial againstacidic and tryptophan residues adjacent to the N-terminal glutaminyl residueand a lack of importance of chain length following the second residue. QPCTcatalyzes N-terminal pyroglutamate formation, also in vitro, it catalyzespyroglutamate formation of N-terminally truncated form of APP amyloid-betapeptides [Glu-3]-beta-amyloid.
QPCT produced in Sf9 Baculovirus cells is a single, glycosylated polypeptide chain containing 339 amino acids (29-361a.a.) and having a molecular mass of 38.7kDa (Molecular size on SDS-PAGE will appear at approximately 28-40kDa). QPCT is expressed with a 6 amino acid His tag at C-Terminus and purified by proprietary chromatographic techniques.
