Progastricsin-C(PGC) is an aspartic proteinase which is synthesized in the gastric mucosa asinactive precursors. PGC is a part of the peptidase family A1 and contains aprosegment which is responsible for stabilizing the inactive form and preventingthe entrance of the substrate to the active site. PGC is used as a biomarkerfor various gastric diseases including Helicobacter pylori related gastritis. PGCis also hydrolyzes various proteins.
PGC produced in Sf9 Baculovirus cells is a single,glycosylated polypeptide chain containing 380 amino acids (17-388 a.a.) andhaving a molecular mass of 41.6kDa (Molecular size on SDS-PAGE will appear atapproximately 40-57kDa).�PGC is expressed with an 8 amino acid His tag atC-Terminus and purified by proprietary chromatographic techniques.

The encoded protein is a digestive enzyme that is produced in the stomach and constitutes a major component of the gastric mucosa. This protein is also secreted into the serum. This protein is synthesized as an inactive zymogen that includes a highly basic prosegment. This enzyme is converted into its active mature form at low pH by sequential cleavage of the prosegment that is carried out by the enzyme itself. Polymorphisms in this gene are associated with susceptibility to gastric cancers. Serum levels of this enzyme are used as a biomarker for certain gastric diseases including Helicobacter pylori related gastritis. Alternate splicing results in multiple transcript variants.
