Human PDE6D was initially identified as a fourth subunit of rod-specific cGMP phosphodiesterase, PDE6. PDE6D is an Oligomer composed of two catalytic chains (alpha and beta), an inhibitory chain (gamma) and the delta chain. PDE6D interacts with RPGR, ARL2 and ARL3. The catalytically active PDE6 is a heterodimer (?Beta) that is controlled by two inhibitory ? subunits. Since PDE6D does not change the catalytic properties of PDE?Beta, its function is still unanswered. PDE6D is expressed specifically in the retina.
PDE6D produced in E.Coli is a single, non-glycosylated polypeptide chain containing 158 amino acids (1-150 a.a.) and having a molecular mass of 18.4kDa. PDE6D is fused to an 8 amino acids His Tag at C-terminus and purified by proprietary chromatographic techniques.
UniProt Protein Function: | PDE6D: Acts as a GTP specific dissociation inhibitor (GDI). Increases the affinity of ARL3 for GTP by several orders of magnitude and does so by decreasing the nucleotide dissociation rate. Stabilizes Arl3-GTP by decreasing the nucleotide dissociation. Belongs to the PDE6D/unc-119 family. |
UniProt Protein Details: | Chromosomal Location of Human Ortholog: 2q35-q36 Cellular Component: cytoplasmic vesicle; cytosol Molecular Function:GTPase inhibitor activity; protein binding; Rab GTPase binding Disease: Joubert Syndrome 22 |
NCBI Summary: | This gene encodes the delta subunit of rod-specific photoreceptor phosphodiesterase (PDE), a key enzyme in the phototransduction cascade. A similar protein in cow functions in solubilizing membrane-bound PDE. In addition to its role in the PDE complex, the encoded protein is thought to bind to prenyl groups of proteins to target them to subcellular organelles called cilia. Mutations in this gene are associated with Joubert syndrome-22. Alternative splicing results in multiple splice variants. [provided by RefSeq, Mar 2014] |
