Description
GNAS Antibody (PACO57068)
The GNAS Polyclonal Antibody (PACO57068) is a versatile tool for researchers studying GNAS, a key regulator of various signaling pathways in cells. This antibody, produced in rabbits, shows high reactivity with human samples and is validated for use in Western blot applications. By specifically binding to the GNAS protein, this antibody enables the detection and analysis of GNAS in different cell types, making it an ideal choice for studies in molecular biology and signal transduction research.GNAS, also known as the guanine nucleotide-binding protein alpha stimulating activity polypeptide, plays a crucial role in mediating the effects of various hormones, neurotransmitters, and other extracellular signals on cellular functions.
Dysregulation of GNAS signaling has been linked to several diseases, including endocrine disorders and certain types of tumors. Therefore, investigating the function and regulation of GNAS is essential for gaining insights into disease mechanisms and potential therapeutic targets. With its high specificity and sensitivity, the GNAS Polyclonal Antibody (PACO57068) provides researchers with a valuable tool for exploring the role of GNAS in normal cellular processes and disease states. Whether studying molecular pathways or potential drug targets, this antibody offers reliable performance and accurate detection, facilitating deeper insights into the complex mechanisms governed by GNAS.

Western Blot. Positive WB detected in: MCF-7 whole cell lysate, HepG2 whole cell lysate. All lanes: GNAS antibody at 3.4µg/ml. Secondary. Goat polyclonal to rabbit IgG at 1/50000 dilution. Predicted band size: 68 kDa. Observed band size: 68 kDa.
IHC image of PACO57068 diluted at 1:600 and staining in paraffin-embedded human brain tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
Immunofluorescence staining of MCF-7 cells with PACO57068 at 1:200, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
Background: | May inhibit the adenylyl cyclase-stimulating activity of guanine nucleotide-binding protein G(s) subunit α which is produced from the same locus in a different open reading frame. |
Synonyms: | Protein ALEX (Alternative gene product encoded by XL-exon), GNAS, GNAS1 |
UniProt Protein Function: | G-alpha(s) iso7: a guanine nucleotide-binding protein of the G12 class of G-alpha proteins. Agonist binding to Gq-coupled receptors may block Akt activation via the release of active G-alpha(q) subunits that inhibit phosphatidylinositol 3-kinase. Heterotrimeric G proteins are composed of 3 units; alpha, beta and gamma. The alpha chain contains the guanine nucleotide binding site. |

NCBI Summary: | This locus has a highly complex imprinted expression pattern. It gives rise to maternally, paternally, and biallelically expressed transcripts that are derived from four alternative promoters and 5' exons. Some transcripts contain a differentially methylated region (DMR) at their 5' exons, and this DMR is commonly found in imprinted genes and correlates with transcript expression. An antisense transcript is produced from an overlapping locus on the opposite strand. One of the transcripts produced from this locus, and the antisense transcript, are paternally expressed noncoding RNAs, and may regulate imprinting in this region. In addition, one of the transcripts contains a second overlapping ORF, which encodes a structurally unrelated protein - Alex. Alternative splicing of downstream exons is also observed, which results in different forms of the stimulatory G-protein alpha subunit, a key element of the classical signal transduction pathway linking receptor-ligand interactions with the activation of adenylyl cyclase and a variety of cellular reponses. Multiple transcript variants encoding different isoforms have been found for this gene. Mutations in this gene result in pseudohypoparathyroidism type 1a, pseudohypoparathyroidism type 1b, Albright hereditary osteodystrophy, pseudopseudohypoparathyroidism, McCune-Albright syndrome, progressive osseus heteroplasia, polyostotic fibrous dysplasia of bone, and some pituitary tumors.
