Description
DIO2 Antibody (PACO58292)
The Dio2 Polyclonal Antibody (PAC058292) is a versatile tool designed for researchers studying the Dio2 protein, which plays a key role in thyroid hormone regulation and metabolism. This antibody, produced using rabbits, exhibits high reactivity with human samples and is validated for use in Western blot applications.Dio2 is an important enzyme responsible for converting the inactive form of thyroid hormone into its active form, impacting various physiological processes such as metabolism, growth, and development.
Dysregulation of Dio2 has been associated with thyroid disorders, metabolic diseases, and cancer.By targeting the Dio2 protein, researchers can investigate its expression levels and activity in different cell types and tissues, providing insights into thyroid hormone signaling pathways and potential therapeutic targets. The Dio2 Polyclonal Antibody is a valuable tool for studies in endocrinology, metabolism, and cancer research.

IHC image of PACO58292 diluted at 1:400 and staining in paraffin-embedded human skeletal muscle tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system. | |
Immunofluorescence staining of A549 cells with PACO58292 at 1:133, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L). |
Background: | Responsible for the deiodination of T4 (3,5,3',5'-tetraiodothyronine) into T3 (3,5,3'-triiodothyronine). Essential for providing the brain with appropriate levels of T3 during the critical period of development. |
Synonyms: | Type II iodothyronine deiodinase (EC 1.21.99.4) (5DII) (DIOII) (Type 2 DI) (Type-II 5'-deiodinase), DIO2, ITDI2 TXDI2 |
UniProt Protein Function: | DIO2: Responsible for the deiodination of T4 (3,5,3',5'- tetraiodothyronine) into T3 (3,5,3'-triiodothyronine). Essential for providing the brain with appropriate levels of T3 during the critical period of development. Belongs to the iodothyronine deiodinase family. 2 isoforms of the human protein are produced by alternative splicing. |

NCBI Summary: | The protein encoded by this gene belongs to the iodothyronine deiodinase family. It catalyzes the conversion of prohormone thyroxine (3,5,3',5'-tetraiodothyronine, T4) to the bioactive thyroid hormone (3,5,3'-triiodothyronine, T3) by outer ring 5'-deiodination. This gene is widely expressed, including in thyroid, placenta, pituitary and brain. It is thought to be responsible for the 'local' production of T3, and thus important in influencing thyroid hormone action in these tissues. It has also been reported to be highly expressed in thyroids of patients with Graves disease, and in follicular adenomas. The intrathyroidal T4 to T3 conversion by this enzyme may contribute significantly to the relative increase in thyroidal T3 production in these patients. This protein is a selenoprotein containing the rare selenocysteine (Sec) amino acid at its active site, and may contain additional Sec residues. Sec is encoded by the UGA codon, which normally signals translation termination. The 3' UTRs of selenoprotein mRNAs contain a conserved stem-loop structure, designated the Sec insertion sequence (SECIS) element, that is necessary for the recognition of UGA as a Sec codon, rather than as a stop signal. Alternatively spliced transcript variants have been described for this gene. [provided by RefSeq, May 2016] |
