Description
CRMP2 (Phospho-Thr514)Colorimetric Cell-Based ELISA Kit
The CRMP2 (Phospho-Thr514) Colorimetric Cell-Based ELISA Kit is a cutting-edge tool designed for the precise measurement of CRMP2 phosphorylation levels in cell lysates. This kit offers exceptional sensitivity and accuracy, allowing researchers to obtain reliable and consistent results for their studies.CRMP2, also known as collapsin response mediator protein 2, is a key signaling molecule involved in various cellular processes such as axon guidance, neurite outgrowth, and neuronal development. Phosphorylation of CRMP2 at Thr514 site is a crucial regulatory mechanism that controls its function and localization within the cell.
This ELISA kit enables researchers to investigate the role of CRMP2 phosphorylation in different cellular pathways and disease conditions, particularly in neurodegenerative disorders, neuronal injuries, and cancer progression. By accurately quantifying CRMP2 phosphorylation levels, scientists can gain valuable insights into the molecular mechanisms underlying these conditions and develop novel therapeutic strategies.

The CRMP2 (Phospho-Thr514) Colorimetric Cell-Based ELISA Kit is a convenient, lysate-free, high throughput and sensitive assay kit that can detect CRMP2 protein phosphorylation and expression profile in cells. The kit can be used for measuring the relative amounts of phosphorylated CRMP2 in cultured cells as well as screening for the effects that various treatments, inhibitors (ie. siRNA or chemicals), or activators have on CRMP2 phosphorylation.
Qualitative determination of CRMP2 (Phospho-Thr514) concentration is achieved by an indirect ELISA format. In essence, CRMP2 (Phospho-Thr514) is captured by CRMP2 (Phospho-Thr514)-specific primary antibodies while the HRP-conjugated secondary antibodies bind the Fc region of the primary antibody. Through this binding, the HRP enzyme conjugated to the secondary antibody can catalyze a colorimetric reaction upon substrate addition. Due to the qualitative nature of the Cell-Based ELISA, multiple normalization methods are needed:
1. | A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalizing the target absorbance values. |
2. | Following the colorimetric measurement of HRP activity via substrate addition, the Crystal Violet whole-cell staining method may be used to determine cell density. After staining, the results can be analysed by normalizing the absorbance values to cell amounts, by which the plating difference can be adjusted. |

NCBI Summary: | This gene encodes a member of the collapsin response mediator protein family. Collapsin response mediator proteins form homo- and hetero-tetramers and facilitate neuron guidance, growth and polarity. The encoded protein promotes microtubule assembly and is required for Sema3A-mediated growth cone collapse, and also plays a role in synaptic signaling through interactions with calcium channels. This gene has been implicated in multiple neurological disorders, and hyperphosphorylation of the encoded protein may play a key role in the development of Alzheimer's disease. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Sep 2011] |

Step | Procedure |
1. | Seed 200 µL of 20,000 adherent cells in culture medium in each well of a 96-well plate. The plates included in the kit are sterile and treated for cell culture. For suspension cells and loosely attached cells, coat the plates with 100 µL of 10 µg/ml Poly-L-Lysine (not included) to each well of a 96-well plate for 30 minutes at 37°C prior to adding cells. |
